Gram stain is used to classify bacteria into two broad groups. These are the gram-positive and the gram-negative bacteria. Practically, the stain differentiates the bacteria by their cell wall chemical and physical properties. Gram positive bacterial cells generally have a thick layer of peptidoglycans in their cell wall. This layer is able to retain the primary stain component in the Gram stain, the crystal violet dye. Gram-negative bacteria, on the other hand, have a thinner or absent peptidoglycan layer that allows the crystal violet dye to wash out on addition of ethanol. Therefore, they stain pink when a counterstain such as safranin is added.
Most antibiotics are either bactericidal or bacteriostatic. They work by preventing synthesis of the peptidoglycan layer of the cell wall. This way, bacterial cells leak their cytoplasmic contents and die. The target enzyme for most antibiotics that target cell wall synthesis is the trans-peptidase enzyme that catalyzes the cross-linking of the pentagylcine bridges with the peptidoglycan side chains. This way, there is a formation of a faulty cell membrane.
A carrier is an animal or person that is infected with a pathogen but shows no symptoms related to the disease process of the infection.
