Abstract
Bacteria are among the most ubiquitous organisms on the planet. The management of bacteria requires the understanding of the organisms which come through studies conducted for their characterization. Aseptic techniques are necessary for success in conducting experiments involving microorganisms. The objective of this study was to isolate colonies of individual strains of bacteria from mixed culture. E.coli, Streptocaucus aurea were inoculated in MacConkey Agar, Manitol salt Agar, Nutrient Agar +7.5% NaCl, Blood Agar and Chocolate Agar to observe the behavior of the organisms. Steak plate technique was also used to inoculate agar plate. The results indicated that E.coli and Streptocaucus aurea were seen to form colonies on different forms of media. The media were also seen to be able to perform their differential function. Various colonies were identified in the agar plate showing the presence of various species of bacteria in the culture. Differential, selective media have the capacity to distinguish between the bacterial colonies.
