Problem
Tumorigenesis describes the multiple step process by which normal cells become neoplastic. Normal cells develop traits that change them into tumorigenic and eventually malignant state. Collectively, these acquired traits are called the hallmarks of cancer and are the result of genomic instability which leads to malignant cell structures. DNA replication is a fundamental process in ensuring accurate copying of genetic information and error minimization during this process. In the event that normal DNA replication experiences perturbations, the induced stress on DNA material, called replication stress (DRS), occurs in the early stages of cancer; DRS causes cellular senescence which prohibits malignant growth.
Chemotherapeutic interventions require that curative drugs in this category should target those biological processes that are more crucial to the spread of tumor cells than of normal cells. One such key process is DNA replication and chemotherapy intervenes either by modifying DNA templates or through enzymatic inhibition leading to stalled progression in replication forks.
